The cells were incubated in 1x PBS containing 10 % normal goat serum to block non-specific protein-protein interactions followed by the antibody (ab233054) (1x10⁶ in 100 μl at 0.04 μg/ml (1/12500)) for 30 min on ice. Isotype control antibody (black line) was mouse IgG2a kappa; Alexa Fluor^® 647 used at the same concentration and conditions as the primary antibody. Unlabelled sample (blue line) was also used as a control. Acquisition of >5,000 events were collected using a 40 mW Red laser (640nm) and 670/14 bandpass filter.

Specificity

This antibody binds to some medulloblastoma and neuroblastoma cells and rhabdomyosarcoma and certain other solid tumours. No binding observed to marrow constituents other than platelets and megakaryocytes.

From Jan 2026, QC testing of replenishment batches of this primary conjugate changed. All tested and expected application and reactive species combinations are still covered by our Abcam product promise. However, the replenishment lots are subjected to bioanalytical testing only. Target data

Integrin alpha-IIb/beta-3 is a receptor for fibronectin, fibrinogen, plasminogen, prothrombin, thrombospondin and vitronectin. It recognizes the sequence R-G-D in a wide array of ligands. It recognizes the sequence H-H-L-G-G-G-A-K-Q-A-G-D-V in fibrinogen gamma chain (By similarity). Following activation integrin alpha-IIb/beta-3 brings about platelet/platelet interaction through binding of soluble fibrinogen (PubMed : 9111081). This step leads to rapid platelet aggregation which physically plugs ruptured endothelial cell surface (By similarity).
