Immunocytochemistry/ Immunofluorescence - Alexa Fluor® 647 Anti-COUP TF1 antibody [EPR10841] (AB210038)
ab210038 staining COUP TF1 in HeLa cells. The cells were fixed with 4% formaldehyde (10 min), permeabilized with 0.1% Triton X-100 for 5 minutes and then blocked with 1% BSA/10% normal goat serum/0.3M glycine in 0.1% PBS-Tween for 1h. The cells were then incubated overnight at +4°C with ab210038 at 1/100 dilution (shown in red) and ab195887, Mouse monoclonal to alpha Tubulin (Alexa Fluor® 488), at 1/250 dilution (shown in green). Nuclear DNA was labelled with DAPI (shown in blue). Image was taken with a confocal microscope (Leica-Microsystems, TCS SP8).
This product also gave a positive signal under the same testing conditions in HeLa cells fixed with 100% methanol (5min).

COUP TF1 influences numerous systems by regulating genes involved in neuronal development and organogenesis. This protein is a core component of transcription complexes interacting with other nuclear receptors and co-regulators to modulate its effects. The activity of COUP TF1 also extends to the regulation of neuronal subtype specification and synaptic connectivity signifying its importance in the central nervous system development.

COUP TF1 participates in essential signaling cascades such as the Notch and retinoic acid pathways. It modulates the retinoic acid response by interacting with the retinoic acid receptor (RAR) thereby influencing cell differentiation and growth. Additionally COUP TF1 interacts with the Notch signaling pathway affecting neural lineage decisions and maintenance.
