ab288325 staining HA tag in HEK293T transfected with HA tagged expression contruct cells. The cells were fixed with 4% formaldehyde (10 min), permeabilised with 0.1% Triton x-100 for 5 minutes and then blocked with 1% BSA/10% normal goat serum/0.3M glycine in 0.1% PBS-Tween for 1h. The cells were then incubated overnight at +4°C with ab288325 at 2 µg/ml (shown in red). ab195887, Mouse monoclonal to alpha Tubulin (Alexa Fluor® 488) (shown in green) was used at 2.5 µg/ml for structural counterstaining.

Image was acquired with a confocal microscope (Leica-Microsystems TCS SP8) and a single confocal section is shown.

The cells were fixed with 4 % formaldehyde (10 min) and then permeabilized with 90% Methanol. The cells were then incubated in 1x PBS containing 10 % normal goat serum to block non-specific protein-protein interaction followed by the antibody (ab288325) (1x 106 in 100 μl at 0.1 μg/ml (1/5000)) for 30 min at 22°C.

Binds to sialic acid-containing receptors on the cell surface, bringing about the attachment of the virus particle to the cell. This attachment induces virion internalization of about two third of the virus particles through clathrin-dependent endocytosis and about one third through a clathrin- and caveolin-independent pathway. Plays a major role in the determination of host range restriction and virulence. Class I viral fusion protein. Responsible for penetration of the virus into the cell cytoplasm by mediating the fusion of the membrane of the endocytosed virus particle with the endosomal membrane.
