Immunohistochemical analysis of paraffin-embedded (A) HEK-293T (human tissue labeling Histone H2B (mutated E76Q) with ab317717 at 1/100 (5.0 ug/ml) dilution.

Positive staining on (B) HEK-293T transfected with a human H2BC4 E76Q expression vector containing a His-tag cell pellet. No staining on (A) HEK-293T transfected with a human H2BC4 (WT) expression vector containing a His-tag cell pellet and (C) HEK-293T transfected with an empty vector containing a His-tag cell pellet.The section was incubated with ab317717 for 60 mins at room temperature (shown in red). Nuclear DNA was labeled with DAPI (shown in blue). The section was then mounted using Fluoromount®.The immunostaining was performed on a Leica Biosystems BOND RX instrument.Image was taken with a confocal microscope (Leica-Microsystems, TCS SP8).

Counterstained with DAPI.

Heat mediated antigen retrieval was performed with Tris-EDTA buffer (pH 9.0, Epitope Retrieval Solution2) for 40 mins

Immunofluorescent analysis of 4% Paraformaldehyde-fixed, 0.1% TritonX-100 permeabilized 293T (human embryonic kidney epithelial cell) cells labelling Histone H2B (mutated E76Q) with ab317717 at 1/250 (2.0 ug/ml) dilution (Green).

Confocal image showing positive staining in 293T cells transfected with a human H2BC4 E76Q expression vector containing a myc tag, while showing no staining in 293T cells transfected with a human H2BC4 (WT) expression vector containing a myc tag(shown in magenta). Myc-Tag Mouse mAb (Alexa Fluor® 488) was used to counterstain tubulin at 1/100 0.5ug/ml dilution (Magenta). The Nuclear counterstain was DAPI (Blue).

Histones thereby play a central role in transcription regulation, DNA repair, DNA replication and chromosomal stability. DNA accessibility is regulated via a complex set of post-translational modifications of histones, also called histone code, and nucleosome remodeling.. Has broad antibacterial activity.
