Overlay histogram showing HepG2 cells stained with ab237120 (red line). The cells were fixed with 80 % methanol (5 min) and then permeabilized with 0.1 % PBS-Triton X-100 for 15 min. The cells were then incubated in 1x PBS containing 10 % normal goat serum to block non-specific protein-protein interaction followed by the antibody (ab237120) (1x 106 in 100μl at 1μg/ml (1/500)) for 30 min at 22°C.

Isotype control antibody (black line) was Rabbit IgG (monoclonal) Alexa Fluor® 647 (ab199093) used at the same concentration and conditions as the primary antibody. Unlabelled sample (blue line) was also used as a control.

Acquisition of >5,000 events were collected using a 40 mW Red laser (640nm) and 670/14 bandpass filter.

This antibody gave a positive signal in HepG2 cells fixed with 4 % formaldehyde (10 min) / permeabilized with 0.1 % PBS-Triton X-100 for 15 min used under the same conditions.

ab237120 staining Nectin 2 in HT-29 cells. The cells were fixed with 100% methanol (5 min), permeabilized with 0.1% Triton X-100 for 5 minutes and then blocked with 1% BSA/10% normal goat serum/0.3M glycine in 0.1% PBS-Tween for 1h. The cells were then incubated overnight at +4°C with ab237120 at 1/100 dilution (shown in red) and ab195887, Mouse monoclonal to alpha Tubulin (Alexa Fluor® 488), at 1/250 dilution (shown in green). Nuclear DNA was labeled with DAPI (shown in blue).

Image was taken with a confocal microscope (Leica-Microsystems, TCS SP8).

This product also gave a positive signal under the same testing conditions in HT-29 cells fixed with 4% formaldehyde (10 min).

Nectin 2 also known as CD112 is a transmembrane cell adhesion molecule belonging to the immunoglobulin superfamily. It plays a critical role in mediating cell-cell adhesion by forming homophilic and heterophilic interactions with other nectins. The protein has a mass of approximately 65 kDa and shows high expression in epithelial and endothelial cells. Nectin 2 has been identified in various tissues including the skin lung and kidney reflecting its importance in maintaining tissue integrity.
Biological function summary

Nectin 2 facilitates cell adhesion and communication playing an important role in the formation of adherens junctions. It serves as part of a larger biological complex with proteins such as afadin linking it to the actin cytoskeleton. Through these interactions Nectin 2 influences the signaling pathways that regulate tissue development and maintenance. Its ability to bind herpes virus entry mediator A (HVEM) further demonstrates its function in viral entry highlighting its diverse biological roles.

Pathways

Nectin 2 participates in the cell adhesion and immune response pathways. It works closely with Nectin 1 and Cadherins within these pathways to mediate cell-cell junction formation and signaling. Nectin 2 relates to cancer and viral infections. Through its role as a herpesvirus entry receptor it also connects to herpes simplex virus (HSV) infections. The interactions between Nectin 2 and other nectins such as Nectin 3 may affect cell proliferation and viral entry expanding the understanding of its involvement in disease mechanisms.

Target data

Modulator of T-cell signaling. Can be either a costimulator of T-cell function, or a coinhibitor, depending on the receptor it binds to. Upon binding to CD226, stimulates T-cell proliferation and cytokine production, including that of IL2, IL5, IL10, IL13, and IFNG. Upon interaction with PVRIG, inhibits T-cell proliferation.
