Immunocytochemistry/ Immunofluorescence - Alexa Fluor® 647 Anti-PKC beta 2 antibody [Y125] (AB195252)
ab195252 staining PKC beta 2 in HeLa cells. The cells were fixed with 100% methanol (5 min), permeabilized in 0.1% Triton X-100 for 5 minutes and then blocked in 1% BSA/10% normal goat serum/0.3M glycine in 0.1%PBS-Tween for 1h. The cells were then incubated with ab195252 at a working dilution of 1 in 100 (shown in red) and ab195887, Mouse monoclonal [DM1A] to alpha Tubulin (Alexa Fluor® 488, shown in green) at 2μg/ml overnight at +4°C. Nuclear DNA was labelled in blue with DAPI.
This product gave a positive signal in 4% formaldehyde (10 min) fixed HeLa cells under the same testing conditions.
Image was taken with a confocal microscope (Leica-Microsystems, TCS SP8).

PKC beta 2 acts as an important regulator in the immune system by modulating immune cell activation and proliferation. It functions as part of the PKC protein complex which interacts with lipids and calcium as cofactors. This protein has critical influence on processes such as B-cell activation and phagocyte response. It also regulates several transcription factors impacting gene expression critical for the immune system.

PKC beta 2 integrates into multiple signaling pathways including the NF-kB and MAPK pathways. These pathways are essential for transmitting signals from the cell surface to the nucleus affecting gene expression and cell function.
