Activity summary

Syntaxin 3 sometimes known as Stx3 is a protein involved in vesicle trafficking and membrane fusion. It plays a mechanical role by facilitating the fusion of transport vesicles with target membranes an essential step in intracellular transport mechanisms. Syntaxin 3 is a member of the t-SNARE family and has a molecular mass of approximately 33 kDa. This protein expresses mainly in epithelial cells including those in the kidney and lungs as well as certain neuronal tissues indicating its involvement in critical cellular processes in these areas.

Biological function summary

Syntaxin 3 acts in cellular transport by participating in the trans-Golgi network-to-plasma membrane pathway. This protein is part of the SNARE complex a group of proteins that mediates vesicle fusion. By engaging with other SNARE proteins like SNAP-25 and VAMP Syntaxin 3 helps ensure the precise delivery of vesicle contents to the plasma membrane. Its role is important for maintaining cellular homeostasis by regulating how cells communicate and exchange materials with their environment.

Pathways

Syntaxin 3 is an integral component of the exocytosis pathway and is involved in the secretory pathway. In exocytosis Syntaxin 3 associates with proteins such as Munc18 and synaptotagmin which help in vesicle docking and fusion. In the secretory pathway Syntaxin 3 is responsible for directing proteins and lipids to their intended membrane sites thereby influencing cellular communication. These interactions guide the flow of proteins within vesicle transport networks and coordinate various signaling events within the cell.

Associated diseases and disorders

Syntaxin 3 has connections to conditions such as cystic fibrosis and certain types of diabetes. In cystic fibrosis abnormal function of Syntaxin 3 may disrupt normal secretory processes in epithelial cells contributing to enhanced pathology. The protein is associated with the cystic fibrosis transmembrane conductance regulator (CFTR) protein which fails to localize correctly in this disease. In diabetes Syntaxin 3 impacts insulin secretion due to its role in pancreatic beta cells tying it to the disorder through insulin-regulating mechanisms with proteins such as GLUT4.
