Transcription factor which plays a key role in the Hippo signaling pathway, a pathway involved in organ size control and tumor suppression by restricting proliferation and promoting apoptosis. The core of this pathway is composed of a kinase cascade wherein MST1/MST2, in complex with its regulatory protein SAV1, phosphorylates and activates LATS1/2 in complex with its regulatory protein MOB1, which in turn phosphorylates and inactivates YAP1 oncoprotein and WWTR1/TAZ. Acts by mediating gene expression of YAP1 and WWTR1/TAZ, thereby regulating cell proliferation, migration and epithelial mesenchymal transition (EMT) induction. Binds specifically and non-cooperatively to the Sph and GT-IIC 'enhansons' (5'-GTGGAATGT-3') and activates transcription. Binds to the M-CAT motif.

Activity summary

TEAD4 also known as TEF-3 or TEF is a transcription factor that plays a significant mechanical role in gene regulation. It belongs to the TEAD family of proteins. TEAD4 has an approximate mass of 48 kDa and is ubiquitously expressed in various tissues. It interacts with different cofactors to regulate gene expression and influences several cellular processes. The protein's expression is especially noted in cancerous tissues emphasizing its role in cellular growth and proliferation.

Biological function summary

TEAD4 is important for embryonic development and cell fate determination. It forms a complex with YAP (Yes-associated protein) to regulate transcriptional activities. The TEAD4-YAP complex binds to DNA driving the expression of genes involved in cell proliferation and survival. TEAD4 also coordinates with _tef_1 and several other transcription factors to regulate genes necessary for development and differentiation influencing processes such as muscle and neural tissue formation.

Pathways
