Claudin18.2 and Claudin18.1 are integral membrane proteins part of the claudin family which are tight junction proteins. Claudin18.2 has a molecular weight of approximately 27 kDa. These proteins are primarily expressed in epithelial cells particularly within the gastric mucosa and lung tissue. They serve as essential components of cell-cell adhesion influencing paracellular transport and maintaining cell polarity.
These proteins are vital in forming tight junction strands contributing to cellular barrier functions. Claudin18.2 and Claudin18.1 belong to a multi-protein complex vital for establishing cell permeability limits. Through their integration in tight junctions they regulate ion passage minimize fluid loss and maintain tissue homeostasis. Their dynamic nature allows adaptation in response to physiological stimuli making them central to epithelial barrier integrity.
Claudins including Claudin18.2 and Claudin18.1 play roles in signaling pathways like Wnt and MAPK which are pivotal for cellular processes like proliferation and differentiation. These pathways involve interactions with proteins like ZO-1 and occludin contributing to the signaling processes across the epithelial cells. This involvement with important signaling proteins highlights their importance in maintaining cell function and signaling responses.
These claudins have strong associations with gastric cancer and asthma pathophysiology. Overexpression of Claudin18.2 has been shown in gastric cancer indicating its potential as a therapeutic target. In asthma altered expression of Claudin18.1 affects lung epithelial integrity linking these proteins to airway inflammation and remodeling. The connection with proteins like E-cadherin and beta-catenin further highlights their role in tumor progression and epithelial transformation making them relevant in clinical research.
