ALG3 also known as alpha-13-mannosyltransferase is an enzyme with a role in the glycosylation process. It is encoded by the ALG3 gene and has a molecular mass of approximately 48 kDa. ALG3 is mainly expressed in the endoplasmic reticulum where it performs its functions in protein modification. This enzyme catalyzes the addition of mannose to lipid-linked oligosaccharide precursors an important step in N-glycan biosynthesis. Its activity and expression are important for the proper folding and quality control of glycoproteins.
Biological function summary

ALG3 functions as an important player in the N-glycosylation pathway. It acts as part of an essential enzymatic complex that modifies the lipid-linked oligosaccharide. This modification is necessary for protein maturation and stability as glycosylation is a post-translational modification influencing protein folding and stability. Dysfunction in this process can result in incorrect protein folding leading to cellular stress or dysfunction.

Dol-P-Man : Man(5)GlcNAc(2)-PP-Dol alpha-1,3-mannosyltransferase that operates in the biosynthetic pathway of dolichol-linked oligosaccharides, the glycan precursors employed in protein asparagine (N)-glycosylation. The assembly of dolichol-linked oligosaccharides begins on the cytosolic side of the endoplasmic reticulum membrane and finishes in its lumen. The sequential addition of sugars to dolichol pyrophosphate produces dolichol-linked oligosaccharides containing fourteen sugars, including two GlcNAcs, nine mannoses and three glucoses. Once assembled, the oligosaccharide is transferred from the lipid to nascent proteins by oligosaccharyltransferases. In the lumen of the endoplasmic reticulum, adds the first dolichyl beta-D-mannosyl phosphate derived mannose in an alpha-1,3 linkage to Man(5)GlcNAc(2)-PP-dolichol to produce Man(6)GlcNAc(2)-PP-dolichol (PubMed : 10581255). Man(6)GlcNAc(2)-PP-dolichol is a substrate for ALG9, the following enzyme in the biosynthetic pathway (PubMed : 10581255).
