This data was developed using ab307156, the same antibody clone in a different buffer formulation. Flow cytometric analysis of 4% paraformaldehyde fixed 90% methanol permeabilized PC-3 (human prostate adenocarcinoma epithelial cell) cells labeling ALKBH3 with ab307156 at 1/50 dilution (1ug) (Red) compared with a Rabbit monoclonal IgG (ab172730) (Black) isotype control and an unlabeled control (cells without incubation with primary antibody and secondary antibody) (Blue). A Goat Anti-Rabbit IgG (Alexa Fluor® 488, ab150081) at 1/2000 dilution was used as the secondary antibody.

This data was developed using ab307156, the same antibody clone in a different buffer formulation. Blocking and diluting buffer and concentration : 5% NFDM/TBST. The identity of the higher MW band at approximately 40 kDa (in lane 4) is unknown. Exposure time : 15 seconds.

Dioxygenase that mediates demethylation of DNA and RNA containing 1-methyladenosine (m1A) (PubMed : 12486230, PubMed : 12594517, PubMed : 16174769, PubMed : 26863196, PubMed : 26863410). Repairs alkylated DNA containing 1-methyladenosine (m1A) and 3-methylcytosine (m3C) by oxidative demethylation (PubMed : 12486230, PubMed : 12594517, PubMed : 16174769, PubMed : 25944111). Has a strong preference for single-stranded DNA (PubMed : 12486230, PubMed : 12594517, PubMed : 16174769, PubMed : 20714506). Able to process alkylated m3C within double-stranded regions via its interaction with ASCC3, which promotes DNA unwinding to generate single-stranded substrate needed for ALKBH3 (PubMed : 22055184). Can repair exocyclic 3,N4-ethenocytosine adducs in single-stranded DNA (PubMed : 25797601).
