ALKBH4 was immunoprecipitated from 0.35 mg of HEK-293 (human epithelial cell line from embryonic kidney) whole cell lysate with ab195379 at 1/30 dilution. Western blot was performed from the immunoprecipitate using ab195379 at 1/1000 dilution. VeriBlot for IP Detection Reagent (HRP) (ab131366), was used for detection at 1/10000 dilution.
Lane 1 : HEK-293 whole cell lysate 10ug (Input).
Lane 2 : ab195379 IP in HEK-293 whole cell lysate.
Lane 3 : Rabbit monoclonal IgG (ab172730) instead of ab195379 in HEK-293 whole cell lysate.
Blocking and dilution buffer and concentration : 5% NFDM/TBST.
Exposure time : 5 seconds.

Target data

Dioxygenase that mediates demethylation of actin monomethylated at 'Lys-84' (K84me1), thereby acting as a regulator of actomyosin-processes (PubMed : 23673617). Demethylation of actin K84me1 is required for maintaining actomyosin dynamics supporting normal cleavage furrow ingression during cytokinesis and cell migration (PubMed : 23673617). In addition to proteins, also demethylates DNA : specifically demethylates DNA methylated on the 6th position of adenine (N(6)-methyladenosine) DNA, thereby regulating Polycomb silencing (By similarity).
