Alpha 1 Acid Glycoprotein also known as AGP or Alpha 1 Glycoprotein is a plasma protein with a molecular weight of approximately 41 to 43 kDa. This protein circulates primarily in the bloodstream and is produced mainly by the liver. AGP is known for its highly glycosylated structure carrying several branched oligosaccharide chains. It exhibits a significant capacity to bind various endogenous and exogenous lipophilic substances influencing their transport and distribution in the body.
Biological function summary

AGP plays a role in modulating the immune response and maintaining homeostasis. As an acute-phase protein its levels rise significantly during inflammation infection and trauma where it acts to modulate the immune system. AGP does not exist as part of a larger protein complex but interacts with plasma lipoproteins and other glycoproteins aiding in its function of maintaining vascular integrity during acute-phase reactions.

Pathways

Alpha 1 Glycoprotein serves as an important component in the inflammatory response and drug-binding pathways. In the inflammatory pathway AGP interacts with cytokines and acute-phase reactants adjusting the body’s inflammatory processes. It is also involved in the binding and distribution of drugs interacting with albumin and other transport proteins to influence the pharmacokinetics of many medications.

AGP levels change in response to conditions like rheumatoid arthritis and liver diseases. In rheumatoid arthritis its increased levels correlate with disease activity and inflammation. In liver diseases the production and plasma concentration of AGP can reflect hepatic synthesis capability. AGP's interaction with cytokines in these conditions often links it to elevated levels of other acute-phase proteins like C-reactive protein reflecting its participation in the pathophysiological processes of inflammation.
