Function

Serine/threonine-protein kinase that detects bacterial pathogen-associated molecular pattern metabolites (PAMPs) and initiates an innate immune response, a critical step for pathogen elimination and engagement of adaptive immunity (By similarity). Specifically recognizes and binds ADP-D-glycero-beta-D-manno-heptose (ADP-Heptose), a potent PAMP present in all Gram-negative and some Gram-positive bacteria (PubMed:30111836). ADP-Heptose-binding stimulates its kinase activity to phosphorylate and activate TIFA, triggering pro-inflammatory NF-kappa-B signaling (By similarity). May be involved in monosodium urate monohydrate (MSU)-induced inflammation by mediating phosphorylation of unconventional myosin MYO9A (By similarity). May also play a role in apical protein transport by mediating phosphorylation of unconventional myosin MYO1A (By similarity). May play a role in ciliogenesis (By similarity).

    ALPK1 Western blot staining using rabbit Anti-ALPK1 antibody

    This data was developed using Anti-ALPK1 antibody [EPR25006-61] ab317807, the same antibody clone in a different buffer formulation.

    In western blot, Anti-ALPK1 antibody [EPR25006-61] ab317807 was shown to bind specifically to ALPK1. Target of interest was observed at 140 kDa in wild-type iBMM cell lysates (lane 1) with no signal observed at this size in ALPK1 knockout cell line (lane 2).

    The identity of the bands below the target band are unknown.

    The wild-type iBMM and ALPK1 knockout iBMM lysates were kindly provided by Dr Feng Shao's lab, NIBS.

    In Western blot, Anti-Vinculin antibody [EPR8185] (Anti-Vinculin antibody [EPR8185] - Loading Control ab129002) staining at 1/10000 dilution.
