Amelotin also known as AMTN is a protein involved in the formation and mineralization of dental enamel. The protein has an estimated mass of approximately 24 kilodaltons. It is expressed in high amounts in ameloblasts which are cells involved in dental enamel production especially in the maturation stage of enamel development.
Biological function summary

The protein encoded by the Amelotin gene plays a significant role in dental epithelial cell differentiation and enamel biomineralization. Amelotin does not appear to form part of a larger protein complex but acts in coordination with other enamel matrix proteins. Its expression increases as ameloblasts transition into maturation stage suggesting it modulates enamel matrix mineralization.

Pathways

The protein influences tooth development processes particularly within the amelogenesis pathway. Amelotin interacts with and complements the roles of other enamel matrix proteins such as amelogenin and enamelin. Its activity contributes to the regulation of calcium and phosphate ion deposition required for enamel hardening.

Malfunction or altered expression of Amelotin associates with amelogenesis imperfecta a disorder affecting enamel development.
