Western blot - Anti-Angiopoietin 4 antibody [EPR24087-66] (AB313126)

ab313126 was shown to specifically react with Angiopoietin-4. Samples were run on an SDS-PAGE gel then transferred onto a nitrocellulose membrane. ab313126 and ab8245 (Mouse anti GAPDH loading control) were incubated overnight at 4°C at 1/1000 and 1/20000 dilutions, respectively. Blots were developed with Goat anti-Rabbit IgG H&L (IRDye® 800CW) preadsorbed and Goat anti-Mouse IgG H&L (IRDye® 680RD) preadsorbed secondary antibodies at 1/20000 dilution for 1 hour at room temperature before imaging.

Human lung tissue lysate at 20 µg with 3% milk in TBS-0.1 % Tween® 20 (TBS-T)

Angiopoietin 4 also known as ANGPT4 or AGP4 is a secreted glycoprotein involved in vascular development. This protein has a molecular mass of around 56 kDa. ANGPT4 is expressed in a variety of tissues including the lung heart and kidney. It functions by interacting with the Tie receptors influencing vascular remodeling and angiogenesis. Angiopoietin 4 shares homology with other angiopoietins but has distinct regulatory roles compared to its counterparts.
Biological function summary

Angiopoietin 4 regulates angiogenesis and vascular permeability. It acts independently and does not part of a known protein complex. ANGPT4 contributes to the restructuring of blood vessels by promoting endothelial cell survival and migration. By balancing the actions of other angiopoietins such as Angiopoietin 1 it supports the maturation and stabilization of vascular networks impacting overall vascular function and growth.

Pathways

Angiopoietin 4 affects early morphogenetic signaling in vascular development. ANGPT4 operates within the angiopoietin-Tie signaling pathway important for angiogenesis and lymphangiogenesis. It interacts with Tie2 a receptor tyrosine kinase to mediate these effects. Within the pathway ANGPT4 coordinates with Angiopoietin 1 and Angiopoietin 2 modulating the balance between vessel stability and growth as required.

Angiopoietin 4 shows relevance to conditions like cancer and diabetic retinopathy. Abnormal expression or signaling of ANGPT4 can lead to defective vascular structures contributing to the pathology of these diseases. In cancer altered ANGPT4 and its interaction with Tie2 may disrupt normal angiogenesis facilitating tumor growth. Additionally in diabetic retinopathy ANGPT4 expression changes can exacerbate abnormal vessel formation highlighting its importance alongside Angiopoietin 2 in disease progression.
