Immunocytochemistry/ Immunofluorescence - Anti-Angiopoietin-like 4/ANGPTL4 antibody [RP23040038] (AB308049)
For immunofluorescence analysis, 70% confluent log phase HUVEC cells treated with 150uM of Cobalt Chloride for 24 h were fixed with 4% paraformaldehyde for 10 minutes, permeabilized with 0.1% Triton X-100 for 10 minutes, and blocked with 2% BSA for 1 hour at room temperature for detection of Angiopoietin-like 4/ANGPTL4 using ab308049 at 2 µg/mL in 0.1% BSA and incubated for 3 hours at room temperature and then labeled with Goat anti-Rabbit IgG (H+L) Secondary Antibody, Alexa Fluor® 488 conjugate at a dilution of 1/2000 for 45 minutes at room temperature (Panel a : green). Nuclei (Panel b : blue) were stained with DAPI F-actin (Panel c : red) was stained with Alexa Fluor® 555 Rhodamine Phalloidin at 1/300 dilution Panel d represents the merged image showing cytoplasmic localization Panel e is untreated cell with no signal Panel f represents control cells with no primary antibody to assess background The images were captured at 60X magnification

What are recombinant multiclonals?
Recombinant multiclonals are a mixture of recombinant antibodies co-expressed from a library of heavy and light chains. They offer several advantages including:
- - The sensitivity of polyclonal antibodies by recognising multiple epitopes
- - High batch-to-batch consistency and reproducibility
- - Improved sensitivity and specificity
- - Long-term security of supply
- - Animal-free batch production

Biological function summary
ANGPTL4 protein influences processes such as lipid metabolism angiogenesis and wound healing. It does not integrate into a large protein complex but functions independently to exert its effects. Inhibition of lipoprotein lipase by ANGPTL4 decreases adipose tissue fat storage and modulates plasma lipid levels. This modulation plays a critical role in maintaining energy balance and glucose homeostasis particularly under metabolic stress conditions.
Pathways
The functionality of ANGPTL4 protein intersects with the PPAR (Peroxisome Proliferator-Activated Receptor) signaling pathway and the glucose homeostasis pathway. It interacts with other proteins such as PPAR-alpha and PPAR-gamma which regulate genes involved in fatty acid oxidation and lipid transport. This interaction influences the body's response to fasting and caloric restriction by altering lipid metabolism and ensuring efficient energy use.
