Coordinates the control of BAF/BANF1 dephosphorylation by inhibiting VRK1 kinase and promoting dephosphorylation of BAF/BANF1 by protein phosphatase 2A (PP2A), thereby facilitating nuclear envelope assembly (PubMed:22770216). May regulate nuclear localization of VRK1 in non-dividing cells (PubMed:31735666). ANKLE2 also known as Ankyrin Repeat And LEM Domain Containing 2 is a protein with an approximate mass of 112 kDa. Mechanically ANKLE2 plays a role in regulating nuclear envelope reconstruction during cell division. It is expressed in various tissues including the brain liver and heart. ANKLE2 locates in the nucleus and cytoplasm highlighting its engagement in cellular functions related to the nuclear envelope and mitosis.
ANKLE2 contributes to maintaining nuclear integrity and proper cell cycle progression. It does not form part of a known complex but interacts closely with other nuclear envelope proteins. ANKLE2 helps in the disassembly and reassembly of the nuclear envelope supporting chromosome segregation and cell division. Its activity ensures the correct formation of nuclear structures post-mitosis preventing potential errors that could disrupt cellular functions.
ANKLE2 is involved in the mitotic spindle checkpoint pathway and cell cycle control pathway. It interacts with proteins like BAF (Barrier to Autointegration Factor) in these pathways which are essential for nuclear envelope reformation. ANKLE2’s functioning in these pathways demonstrates its participation in monitoring and ensuring successful cell cycle transitions particularly during the mitotic phase.
ANKLE2 is associated with microcephaly and other neurodevelopmental disorders. Mutations in ANKLE2 have connections to impaired brain development highlighting its significance in neural progenitor cell division. ANKLE2 interacts with Zika virus proteins which has been linked to microcephaly resulting from viral infection. Further research into ANKLE2 and related proteins could provide insights into preventing and treating these conditions.
