ANKRD2 also known as Arpp and MARP2 is an ankyrin repeat protein with a molecular mass of approximately 36 kDa. It is mainly expressed in skeletal muscle and cardiac muscle tissues. The protein functions by interacting with other proteins contributing to muscle structure and function. ANKRD2 binds to titin a critical sarcomeric protein playing a role in muscle elasticity and integrity.
Biological function summary

ANKRD2 acts as a stretch sensor within muscle fibers. This role allows it to be part of a complex responsible for mediating mechanosignaling pathways that respond to mechanical stress or damage in muscle cells. ANKRD2 interacts with other signaling molecules like the transcription factor PML which modulates gene expression in response to mechanical cues.

Pathways

ANKRD2 participates in the muscle-specific mechanotransduction cascade. It plays a significant role in the modulation of MAPK signaling which is important for muscle growth and regeneration. The protein associates with other important proteins in this pathway such as titin and PML which work together to regulate muscle adaptation to physical stress.

ANKRD2 has an association with cardiomyopathy and muscular dystrophy. Its interactions with proteins such as titin and PML may influence the pathology of these diseases as these proteins are key in maintaining muscle and cardiac tissue stability. Disruption in ANKRD2 function or expression can contribute to muscular and cardiac impairments highlighting its importance in muscle-related conditions.
