Supplementary information

ANKRD28 also known as Ankyrin Repeat Domain 28 is a protein that contains several ankyrin repeat motifs which are known for mediating protein-protein interactions. It has a molecular mass of approximately 205 kDa. ANKRD28 is expressed in various tissues with higher expression observed in the brain and liver. The presence of ankyrin repeat motifs suggests that ANKRD28 plays a role in scaffolding or signaling facilitating the interaction between multiple proteins.
Biological function summary

ANKRD28 functions as a regulatory component that participates in cellular processes by forming complexes with other proteins. It is known to interact with protein phosphatase 6 (PP6) playing a role in signaling pathways and cellular regulation. Proper interaction within these complexes contributes to maintaining cellular homeostasis highlighting ANKRD28's importance in cellular signaling networks.

Pathways

ANKRD28 links to critical cellular mechanisms such as the NF-kB signaling pathway and stress response pathways. Within these pathways ANKRD28 modulates the function of downstream effector molecules including the phosphorylation status of IkB proteins which are essential for the regulation of NF-kB activity. ANKRD28 is related through these pathways to proteins like PP6 an important serine/threonine phosphatase involved in distributing cellular responses to various stimuli.

ANKRD28 has been associated with conditions like cancer and inflammatory diseases. In cancer altered expression or mutation of ANKRD28 can disrupt signaling pathways leading to uncontrolled cell growth and division. In inflammatory disorders dysregulation of NF-kB signaling potentially mediated by ANKRD28 contributes to excessive inflammatory responses. ANKRD28's interaction with proteins like I-kappaB kinase further connects it to disease progression affecting cellular processes that can lead to these conditions.

Target data

Putative regulatory subunit of protein phosphatase 6 (PP6) that may be involved in the recognition of phosphoprotein substrates. Involved in the PP6-mediated dephosphorylation of NFKBIE opposing its degradation in response to TNF-alpha. Selectively inhibits the phosphatase activity of PPP1C. Targets PPP1C to modulate HNRPK phosphorylation.
