Small-conductance calcium-activated nonselective cation (SCAN) channel which acts as a regulator of phospholipid scrambling in platelets, osteoblasts and fetal thymocytes. Phospholipid scrambling results in surface exposure of phosphatidylserine which in platelets is essential to trigger the clotting system whereas in osteoblasts is essential for the deposition of hydroxyapatite during bone mineralization. Has calcium-dependent phospholipid scramblase activity; scrambles phosphatidylserine, phosphatidylcholine and galactosylceramide. Can generate outwardly rectifying chloride channel currents in airway epithelial cells and Jurkat T lymphocytes.

Anoctamin 6 also known as ANO6 TMEM16F or NGEP is a protein that functions mechanically as a calcium-activated chloride channel and scramblase. It plays a role in phospholipid scrambling across the plasma membrane influencing membrane stability and shape. ANO6 has a mass of approximately 114 kDa. Researchers have detected ANO6 in a variety of tissues with higher expression noted in the heart skeletal muscles and testis.
ANO6 is involved in critical cellular processes such as apoptosis and blood coagulation. This protein contributes to the externalization of phosphatidylserine a process necessary during cell death and platelet activation. ANO6 does not strictly function alone; it often interacts with other members of the TMEM16 family forming complexes that modulate its functional specificity and activity.
ANO6 is integral to the extrinsic pathway of apoptosis and the process of blood coagulation. It plays a role in regulating cell membrane ion homeostasis working alongside related proteins such as ANO1 and ANO2. These pathways maintain the stability and balance of cellular environments influencing processes like cell shrinkage and coagulative events.
ANO6 relates to conditions like Scott syndrome a bleeding disorder characterized by defective phosphatidylserine exposure on platelets and can impact certain cardiac arrhythmias. ANO6's role in these disorders often involves interaction with other regulatory proteins such as phospholipase C which impacts phospholipid signaling and platelet function. Understanding ANO6's involvement in these processes aids in developing potential therapeutic strategies.

ANO6 was immunoprecipitated from 0.35 mg mouse spleen lysate with ab234422 at 1/30 dilution. Western blot was performed from the immunoprecipitate using ab234422 at 1/1000 dilution. VeriBlot for IP Detection Reagent (HRP) (VeriBlot for IP Detection Reagent (HRP) ab131366), was used at 1/5000 dilution.
