Immunoprecipitation - Anti-ANO6 antibody [EPR20910-105] - BSA and Azide free (AB256302)

ANO6 was immunoprecipitated from 0.35 mg mouse spleen lysate with ab234422 at 1/30 dilution. Western blot was performed from the immunoprecipitate using ab234422 at 1/1000 dilution. VeriBlot for IP Detection Reagent (HRP) (ab131366), was used at 1/5000 dilution.
Lane 1 : Mouse spleen lysate 10 μg (Input).
Lane 2 : ab234422 IP in mouse spleen lysate.
Lane 3 : Rabbit monoclonal IgG (ab172730) instead of ab234422 in mouse spleen lysate.
Blocking/Dilution buffer : 5% NFDM/TBST.
Exposure time : 3 mins.

The molecular weight observed is consistent with literatures (PMID : 29507235, PMID : 26417084, PMID : 27227820, PMID : 24478309)

This data was developed using the same antibody clone in a different buffer formulation containing PBS, BSA, glycerol, and sodium azide (ab234422).

Anoctamin 6 also known as ANO6 TMEM16F or NGEP is a protein that functions mechanically as a calcium-activated chloride channel and scramblase. It plays a role in phospholipid scrambling across the plasma membrane influencing membrane stability and shape. ANO6 has a mass of approximately 114 kDa. Researchers have detected ANO6 in a variety of tissues with higher expression noted in the heart skeletal muscles and testis.

Target data

Small-conductance calcium-activated nonselective cation (SCAN) channel which acts as a regulator of phospholipid scrambling in platelets, osteoblasts and fetal thymocytes. Phospholipid scrambling results in surface exposure of phosphatidylserine which in platelets is essential to trigger the clotting system whereas in osteoblasts is essential for the deposition of hydroxyapatite during bone mineralization. Has calcium-dependent phospholipid scramblase activity; scrambles phosphatidylserine, phosphatidylcholine and galactosylceramide. Can generate outwardly rectifying chloride channel currents in airway epithelial cells and Jurkat T lymphocytes.
