Anoctamin 5 also known as ANO5 or TMEM16E is a protein that functions as a calcium-activated chloride channel. It plays a role in phospholipid scramblase activity. Anoctamin 5 is a member of the anoctamin family characterized by ten transmembrane domains. The protein has a molecular mass of approximately 114 kDa. It is expressed in various tissues with notable presence in skeletal muscle cardiac muscle and bone.
Biological function summary

Anoctamin 5 contributes to the regulation of muscle membrane integrity and bone mineralization. It does not function in isolation; it associates with other proteins to maintain the structure and function of muscle cells. In skeletal muscle Anoctamin 5 impacts sarcolemmal repair. In bones it influences the mineralization process by regulating the balance of calcium ions. The protein indirectly affects cell adhesion and signaling by modulating ion channel function.

Pathways

Anoctamin 5 engages in the muscle repair and mineralization pathways. It plays a part in the sarcolemmal repair pathway working closely with dysferlin another protein involved in muscle cell membrane repair. Anoctamin 5’s activity in ion transport is important for maintaining ionic homeostasis during muscle stress and repair events. In the bone mineralization pathway it influences the availability of calcium ions affecting bone density and strength.

Anoctamin 5 mutations have been linked to conditions such as limb-girdle muscular dystrophy type 2L (LGMD2L) and Miyoshi muscular dystrophy type 3 (MMD3). These are genetic disorders characterized by progressive muscle weakness due to impaired muscle membrane repair. The relationship between Anoctamin 5 and these disorders highlights its importance in muscle integrity. In these diseases anomalies in Anoctamin 5 expression or function can disrupt pathways shared with dysferlin exacerbating muscle deterioration.
