Antizyme inhibitor 1 (AZIN1) also known as ornithine decarboxylase antizyme inhibitor (OAZI) is a regulatory protein that modulates polyamine levels in the cell. The mass of AZIN1 is approximately 50 kDa. It is a cytoplasmic enzyme and expressed in various tissues including the liver and pancreas. AZIN1 competes with ornithine decarboxylase (ODC) for binding to antizyme which is a negative regulator of ODC. Unlike ODC AZIN1 lacks catalytic activity but plays an important role in cellular regulation.
Biological function summary

AZIN1 significantly influences polyamine biosynthesis by preventing the formation of inhibitory antizyme-ODC complexes. This effectively regulates polyamine levels that are important for cell growth and proliferation. Antizyme inhibitor (AZI) protein that positively regulates ornithine decarboxylase (ODC) activity and polyamine uptake. AZI is an enzymatically inactive ODC homolog that counteracts the negative effect of ODC antizymes (AZs) OAZ1, OAZ2 and OAZ3 on ODC activity by competing with ODC for antizyme-binding (PubMed : 17900240, PubMed : 26305948). Inhibits antizyme-dependent ODC degradation and releases ODC monomers from their inactive complex with antizymes, leading to formation of the catalytically active ODC homodimer and restoring polyamine production (PubMed : 17900240).
