AP-A also known as Aminopeptidase A is an enzyme that plays a critical role in cellular processes by cleaving N-terminal amino acids from peptide substrates. It has a molecular mass of approximately 100 kDa. AP-A is highly expressed in the brain particularly in the hippocampus and glial cells as well as in other tissues like the kidney and placenta. Its activity has been observed in the synaptic membranes signifying its importance in neurobiological functions.
Aminopeptidase A participates in the regulation of neuropeptides and peptide hormones. It forms part of the renin-angiotensin system where it influences blood pressure and fluid balance. By converting angiotensin II to angiotensin III AP-A modulates these physiological processes. Moreover it is involved in degrading enkephalins which impacts pain perception and emotional responses. This enzyme interacts with other aminopeptidases in the body contributing to various cellular functions.
AP-A significantly impacts the renin-angiotensin system and the endothelin pathway. It influences the cardiovascular system by processing angiotensin peptides working closely with proteins such as angiotensin-converting enzyme (ACE). In the endothelin pathway AP-A affects peptide regulation and is associated with the activity of endothelin-converting enzyme. These pathways demonstrate the enzyme's role in cardiovascular health and homeostasis.
Aminopeptidase A is linked to hypertension and neurological disorders like Alzheimer's disease. Increased activity or expression of AP-A is often seen in the context of high blood pressure where it works in conjunction with ACE to modulate blood pressure levels. In Alzheimer's disease altered AP-A activity affects neuropeptide regulation linking it to beta-amyloid accumulation. These associations highlight the enzyme's potential as a target for therapeutic interventions in hypertension and neurodegenerative diseases.
