Supplementary information

AP1S1 also known as Adaptor Protein complex 1 Sigma 1 is an integral component of the AP-1 adaptor complex. This complex is involved in the sorting and trafficking of transmembrane proteins. AP1S1 has a molecular mass of approximately 19 kDa and is universally expressed in various tissues including the liver and brain. Its main function involves the interaction with clathrin aiding in the selection of cargo proteins for transport between the trans-Golgi network and endosomes.
Biological function summary

AP1S1 plays a significant role in vesicular trafficking processes. It participates as part of the AP-1 adaptor protein complex which is responsible for mediating sorting signals in coated vesicles. Through this participation AP1S1 influences the efficiency and specificity of intracellular transport. The AP-1 complex also interacts with phosphatidylinositol phosphates further regulating the vesicle formation and protein sorting mechanisms within cells. This protein's involvement is critical for maintaining cellular homeostasis and specific protein function.

Pathways

AP1S1 functions in pathways related to protein sorting and vesicular transport. It plays a role within the clathrin-mediated endocytosis pathway important for cellular receptor regulation and nutrient uptake. AP1S1 connects with other proteins such as clathrin and other adaptor proteins to form a network ensuring efficient cargo selection and transportation. This function ensures a streamlined process for the cellular uptake and regulation of essential molecules.

AP1S1 has links to various conditions through its role in protein trafficking. Mutations in AP1S1 associate with Mednik Syndrome a disorder characterized by neurological issues due to improper protein sorting. Another connection involves its association with hereditary spastic paraplegia where misregulation of protein trafficking results in neurological deterioration. In these contexts AP1S1 interacts with proteins like AP1B1 which similarly impacts vesicular transport and sorting pathways affecting overall cellular functionality.

Target data

Subunit of clathrin-associated adaptor protein complex 1 that plays a role in protein sorting in the late-Golgi/trans-Golgi network (TGN) and/or endosomes. The AP complexes mediate both the recruitment of clathrin to membranes and the recognition of sorting signals within the cytosolic tails of transmembrane cargo molecules.
