AP3 also known as adaptor-related protein complex 3 is a part of the adaptor protein (AP) complex family involved in cargo selection and vesicle formation for intracellular transport. The complex consists of four subunits with a combined molecular mass of approximately 230 kDa. AP3 predominantly expresses in brain lungs and skin tissues. As a cytosolic complex it helps in the trafficking of proteins to intracellular organelles specifically lysosomes and lysosome-related organelles.
Biological function summary

AP3 plays a central role in the targeting and transport of membrane proteins acting as a scaffold assembling different molecular cargo. It is a component of the AP complex involved in vesicular trafficking within the cell. AP3 interacts with the cytoplasmic tails of cargo proteins to mediate the sorting from the trans-Golgi network to endosomal-lysosomal system. Its function ensures proper protein distribution critical for cellular homeostasis and specialized cellular functions.

Pathways

AP3 influences intracellular trafficking pathways essential for protein sorting. It is especially significant in the biogenesis of lysosome-related organelles. This function aligns it closely with other proteins such as AP1 and AP2 showcasing interconnected roles in protein and membrane trafficking systems. In particular AP3 shares pathways with proteins associated with clathrin contributing to the sorting of integral membrane proteins in endosomal-lysosomal traffic.

Mutations or dysfunction of AP3 are connected to Hermansky-Pudlak syndrome type 2 and other related lysosomal storage disorders. Such ailments result from defective protein sorting and trafficking leading to improper organelle formation and function. In the context of these conditions AP3's interaction with lysosomal proteins becomes important for maintaining normal cellular operations. Moreover AP3 shares pathogenic connectivity with proteins like BLOC-1 and VPS33 highlighting the complexities in these hereditary syndromes.

Target data

Part of the AP-3 complex, an adaptor-related complex which is not clathrin-associated. The complex is associated with the Golgi region as well as more peripheral structures. It facilitates the budding of vesicles from the Golgi membrane and may be directly involved in trafficking to lysosomes. Involved in process of CD8+ T-cell and NK cell degranulation (PubMed : 26744459). In concert with the BLOC-1 complex, AP-3 is required to target cargos into vesicles assembled at cell bodies for delivery into neurites and nerve terminals (By similarity).
