Transmembrane Activator and CAML Interactor known as TACI also goes by the names TNFRSF13B and CD267. TACI is a receptor with a molecular weight of approximately 33 kDa. It is expressed on B cells a type of lymphocyte and to some extent on activated T cells. This receptor plays a part in the immune system helping manage the production and function of certain antibodies.
Biological function summary

TACI involves the regulation of B cell function and survival. It binds to two ligands APRIL and BAFF and TACI is not part of any larger complex. Upon ligand binding TACI can promote antibody class-switching which adjusts the type of antibody that B cells produce. This action supports the immune response by allowing the body to adapt to different pathogens effectively.

Pathways

Immune responses involve TACI in the BAFF-APRIL signaling pathway. The interplay between TACI and its ligands influences the NF-kB signaling cascade leading to B cell proliferation and survival. TACI interacts with other proteins such as BCMA and BAFF-R which are important in regulating B cell homeostasis and function ensuring balanced and effective immune responses.

Aberrant TACI function contributes to conditions like Common Variable Immunodeficiency (CVID) and Systemic Lupus Erythematosus (SLE). In CVID mutations in the TACI gene may lead to impaired antibody production resulting in frequent infections. In the context of SLE TACI's interactions with BAFF and APRIL can contribute to the overactivation of B cells causing autoimmune characteristics of the disease. TACI's role in these conditions makes it a focus for studying therapeutic interventions.
