APC15 also known as Anaphase Promoting Complex subunit 15 is an important regulatory component of the anaphase-promoting complex/cyclosome (APC/C). This protein with a molecular mass of around 16 kDa plays an important role in cell cycle regulation. APC15 expression occurs in various tissues where it contributes to the timely progression of the cell cycle by controlling the degradation of specific cell cycle regulators. It functions mechanistically by recognizing and targeting substrates for ubiquitination which marks them for degradation by the proteasome.
Biological function summary

APC15 contributes to the molecular machinery that drives cell cycle transitions. It is part of the larger APC/C complex a multi-subunit E3 ubiquitin ligase complex essential for maintaining the proper sequence of cell cycle events particularly during mitosis. APC15's regulatory activity ensures that cells do not prematurely enter anaphase enabling the correct chromosomal alignment and segregation. It interacts with other complex subunits to coordinate precise ubiquitination of proteins that inhibit chromosome separation until the cellular environment is ready.

Pathways

APC15 plays a significant role in cell cycle pathways particularly the cell cycle G2/M transition and mitotic exit. It works closely with other proteins like CDC20 and CDH1 which serve as co-activators to manage progression through the cell cycle. These interactions help direct the APC/C complex to its specific substrates at various stages ensuring fidelity in chromosome segregation and the prevention of aneuploidy a condition with too many or too few chromosomes.

APC15 has a connection to cancer where dysregulation can lead to abnormal cell division and tumorigenesis. This dysregulation often involves overexpression or malfunctioning of associated proteins such as CDC20 impacting the APC/C complex's ability to perform adequate protein degradation and cell cycle control. Additionally defects in the APC/C machinery which includes APC15 have been implicated in neurodegenerative diseases where aberrant protein accumulation and misregulation disrupt normal cellular function.

Component of the anaphase promoting complex/cyclosome (APC/C), a cell cycle-regulated E3 ubiquitin ligase that controls progression through mitosis and the G1 phase of the cell cycle (PubMed : 21926987). The APC/C complex catalyzes assembly of branched 'Lys-11'-/'Lys-48'-linked branched ubiquitin chains on target proteins (PubMed : 29033132). In the complex, plays a role in the release of the mitotic checkpoint complex (MCC) from the APC/C : not required for APC/C activity itself, but promotes the turnover of CDC20 and MCC on the APC/C, thereby participating in the responsiveness of the spindle assembly checkpoint (PubMed : 21926987). Also required for degradation of CDC20 (PubMed : 21926987).
