APCDD1 also known as Adenomatosis Polyposis Coli Down-Regulated 1 is a membrane-bound protein involved in the regulation of cell proliferation and differentiation. APCDD1 has a molecular weight of approximately 42 kDa and is predominantly expressed in hair follicles but also found in other tissues such as skin and brain. Its expression often correlates with developmental stages indicating its role in cell growth processes.
Biological function summary

APCDD1 regulates cellular signaling by inhibiting Wnt/β-catenin signaling pathway an important pathway involved in cell fate determination and proliferation. APCDD1 does not function as a classic receptor or kinase but it modulates activity by interacting with other components of this pathway. It is often implicated in controlling the balance between cell proliferation and differentiation particularly in skin tissue.

Pathways

The Wnt/β-catenin signaling pathway features importantly in APCDD1's activities. This pathway critical for regulating embryonic development and adult tissue homeostasis involves interaction with proteins such as β-catenin and LRP5/6 receptors. APCDD1's inhibitory effects contribute to the modulation of these downstream signaling events linking it closely with the regulation of cellular growth and preventing excessive proliferation.

Hair loss conditions such as hereditary hypotrichosis simplex relate to APCDD1 due to its role in follicle development. Mutations or dysregulation of APCDD1 interrupt normal Wnt pathway inhibition leading to altered hair growth cycles. Additionally colorectal cancer is associated with changes in APCDD1 activity where aberrant Wnt signaling plays a role in tumorigenesis. In these diseases proteins like APC and β-catenin become central to the pathological processes influenced by APCDD1 activity.
