Western blot analysis was performed on membrane enriched extracts (30 µg of lysate) of HepG2 (Lane 1), NTERA-2 (Lane 2), HEL.92.1.7 (Lane 3), K562 (Lane 4), HeLa (Lane 8) and tissue extracts of Mouse Liver (Lane 5), Mouse Testis (Lane 6) Rat Testis (Lane 7) and Mouse Lung (Lane 9). The blots were probed with ab308018 at 1-2 µg/mL dilution and detected by chemiluminescence using Goat anti-Rabbit IgG (H+L) Secondary Antibody, HRP conjugate at 1/2500 dilution. A 43 kDa band corresponding to APLNR protein was observed. Known quantity of protein samples were electrophoresed using a 4-12% Bis-Tris gel, electrophoresis system and pre-stained protein standard. Resolved proteins were then transferred onto a nitrocellulose membrane. The membrane was probed with the relevant primary and secondary Antibody following blocking with 5% skimmed milk. Chemiluminescent detection was performed (ECL).

The APJ Receptor also known as the Apelin Receptor (APLNR) is a G protein-coupled receptor with a molecular mass of approximately 42 kDa. You find APJ Receptor highly expressed in tissues like the brain heart and lungs. It acts as a receptor for the peptides called apelin and elabela playing a critical role in signaling processes across various tissues. The receptor demonstrates complex mechanical actions by coupling with G proteins to inhibit adenylate cyclase activity and regulate other intracellular signaling pathways.
