Immunohistochemistry (Formalin/PFA-fixed paraffin-embedded sections) analysis of human cerebrum tissue sections labeling APLP2 with purified ab140624 at 1 : 500 (0.188 μg/ml). Heat mediated antigen retrieval was performed using Bond™ Epitope Retrieval Solution 2 (pH 9.0). Tissue was counterstained with Hematoxylin. LeicaDS9800 (Bond™ Polymer Refine Detection) was used. PBS instead of the primary antibody was used as the negative control. The immunostaining was performed on a Leica Biosystems BOND® RX instrument.

APLP2 also known as Amyloid Precursor-Like Protein 2 is a cell surface protein with a mass of about 92 kDa. This protein belongs to the same family as Amyloid Precursor Protein (APP). APLP2 is notably expressed in many tissues including the brain where its expression is significant. It participates in a variety of cellular functions due to its structural and functional properties similar to that of other amyloid precursor proteins.
Biological function summary

APLP2 participates in the regulation of neuronal growth and synaptic stability. It functions as a part of a larger protein complex and shares structural similarities with other members of its family which suggests overlapping functions. APLP2 interacts with extracellular matrix proteins and may help with cell adhesion processes which are important for maintaining tissue structure and function.

Pathways

The APLP2 protein interacts within signaling pathways that regulate neural development and cell viability. It plays a role in the Notch signaling pathway which is important for cell differentiation processes. Other related proteins such as APP and APLP1 demonstrate similar roles within these pathways indicating a potential coordination among these family members to achieve functional outcomes in tissue development and homeostasis.

APLP2 has been implicated in neurodegenerative diseases such as Alzheimer's disease through its relation to APP. Elevated or misregulated levels of APLP2 have shown connections to increased amyloid-beta production which is a hallmark of Alzheimer's pathology. Furthermore APLP2's involvement in tumorigenesis links it to gliomas with its interaction with proteins like EGFR hinting at its role in cancer-related cellular signaling.
