Supplementary information

APOPT1 also known as Apo-T1 or CCDC86 is a protein involved in the regulation of mitochondrial function. It has a molecular mass of approximately 22 kDa. APOPT1 is mainly expressed in the tissues with high energy demands such as the heart and liver making it significant for energy metabolism. The protein functions within the mitochondria where it participates in the regulation of oxidative phosphorylation and influences mitochondrial integrity.
Biological function summary

The protein plays a significant role in protecting cells from oxidative stress by maintaining the proper function of the electron transport chain. APOPT1 is not part of a larger complex but acts as a solitary operator within mitochondrial pathways. It helps ensure that energy production remains efficient preventing cellular damage through the accumulation of reactive oxygen species. This protective mechanism supports the survival of cells under stress conditions.

Pathways

APOPT1 has a notable role in the apoptosis and mitochondrial biogenesis pathways. It interacts with proteins like Cytochrome c (CYCS) and Apoptotic protease activating factor 1 (APAF1) which are important in the intrinsic pathway of apoptosis. APOPT1's activity within the mitochondrial biogenesis pathway supports the creation and maintenance of new mitochondria which is essential for energy balance and cellular health.

APOPT1 is linked to mitochondrial disorders particularly those affecting the nervous and muscular systems. These disorders often lead to symptoms such as muscle weakness and neurodegeneration. The protein has connections to Cytochrome c oxidase deficiency a condition causing impaired electron transport chain function. Studies suggest that mutations or dysfunctions in APOPT1 can lead to improved understanding of the pathogenesis and potential therapeutic targets for these diseases.
