Supplementary information

Aquaporin-6 (AQP-6) is an integral membrane protein and part of the aquaporin family primarily involved in water transport across cell membranes. It weighs approximately 28 kilodaltons. Unlike other aquaporins AQP-6 also allows passage of anions making it unique in its function within the family. AQP-6 is expressed mainly in kidney tissues particularly in renal collecting ducts where it facilitates fluid balance and ion exchange.
Biological function summary

This protein plays a role in maintaining osmotic balance and modulating cell volume. AQP-6 interacts with other membrane proteins but it is not typically part of larger macromolecular complexes. Its activity contributes significantly to renal urine concentration influencing water reabsorption and ion transport in the kidneys. AQP-6 ion permeability distinguishes it from other aquaporins allowing it to assist in the rapid response to cellular pH changes and osmotic shifts.

Pathways

AQP-6 is involved in the renal water reabsorption and acid-base balance pathways. It interacts with proteins such as AQP-1 and AQP-2 which are also prominent in water transport systems. AQP-6 affects physiological processes governed by hormonal regulation responding to vasopressin to modify water reabsorption rates accordingly.

Abnormalities in AQP-6 function or expression can relate to conditions like nephrogenic diabetes insipidus and acid-base disturbances. Dysfunctional AQP-6 impacts water balance and pH regulation leading to potential kidney-related disorders. The protein's interaction with AQP-2 in water reabsorption processes suggests a possible compensatory mechanism in these pathological states.
