Multiplex immunohistochemistry analysis of formalin/PFA-fixed paraffin-embedded mouse lung tissue staining CD62L with ab325322 at a 1/1000 (0.509 μg/ml) dilution, ab305303 anti-Aquaporin-5 used at a 1/2000 (0.25 μg/ml) dilution and ab324911 anti-ATP-binding cassette sub-family A member 3 used at a 1/2000 (0.253 μg/ml) dilution.

Panel A : anti-CD62L (green; Opal™520), anti-Aquaporin-5 (magenta; Opal™690), anti-ATP-binding cassette sub-family A member 3 (gray; Opal™570) on mouse lung.

The section was incubated in three rounds of staining : in the order of ab325322, ab305303 and ab324911 for 30 mins at room temperature. Each round was followed by a separate fluorescent tyramide signal amplification system.

The immunostaining was performed on a Leica Biosystems BOND® RX instrument with an Opal™ 4-color kit. Image acquisition was performed with Leica SP8 confocal microscope.

Heat mediated antigen retrieval was performed with Tris-EDTA buffer (pH 9.0, Epitope Retrieval Solution2) for 20 mins.

The protein known as Aquaporin 5 often abbreviated as AQP5 is a member of the aquaporin family which are integral membrane proteins that function as channels for water. This particular protein facilitates water transport across cell membranes an action critical in maintaining fluid balance. Aquaporin 5 has a molecular mass of approximately 29 kDa. It is largely expressed in epithelial cells in various tissues including the salivary glands lacrimal glands and respiratory tissues. It is recognized by different names such as Aquaporin-5 and sometimes referenced in studies as Aquazol 5.

Aquaporin 5 is directly associated with conditions such as Sjögren's syndrome and xerostomia (dry mouth syndrome). Aquaporins form homotetrameric transmembrane channels, with each monomer independently mediating water transport across the plasma membrane along its osmotic gradient (By similarity). Plays an important role in fluid secretion in salivary glands (PubMed : 10400615, PubMed : 16571723, PubMed : 18027168). Required for TRPV4 activation by hypotonicity. Together with TRPV4, controls regulatory volume decrease in salivary epithelial cells (PubMed : 16571723). Seems to play a redundant role in water transport in the eye, lung and in sweat glands (PubMed : 10619865, PubMed : 12042359, PubMed : 18027168).
