Multiplex immunohistochemistry analysis of formalin/PFA-fixed paraffin-embedded human lung tissue staining SFTPA1 with ab325321 at a 1 : 1000 (0.512 ug/ml) dilution, ab213363 anti-CD68 used at 1 : 500 (1.314 ug/ml) dilution and ab239904 anti-Aquaporin 5 used at a 1 : 500 (2.0 ug/ml) dilution.

The section was incubated in three rounds of staining : in the order of ab325324, ab213363 and ab239904 for 30 mins at room temperature. Each round was followed by a separate fluorescent tyramide signal amplification system.

The immunostaining was performed on a Leica Biosystems BOND® RX instrument with an Opal™ 4-color kit. Image acquisition was performed with Leica SP8 confocal microscope.

Nuclear counter stain with DAPI.

Heat mediated antigen retrieval with Tris-EDTA buffer (pH 9.0, epitope retrieval solution2) for 20 mins.

Immunohistochemistry (Formalin/PFA-fixed paraffin-embedded sections) analysis of human stomach tissue sections labeling Aquaporin 5 with purified ab92320 at 1/1200 dilution (0.13 μg/ml). Heat mediated antigen retrieval was performed using Perform heat mediated antigen retrieval using ab93684 (Tris/EDTA buffer, pH 9.0). Tissue was counterstained with Hematoxylin. ImmunoHistoProbe one step HRP Polymer (ready to use) secondary antibody was used at 1/0 dilution. PBS instead of the primary antibody was used as the negative control.

The protein known as Aquaporin 5 often abbreviated as AQP5 is a member of the aquaporin family which are integral membrane proteins that function as channels for water. This particular protein facilitates water transport across cell membranes an action critical in maintaining fluid balance. Aquaporin 5 has a molecular mass of approximately 29 kDa. It is largely expressed in epithelial cells in various tissues including the salivary glands lacrimal glands and respiratory tissues. It is recognized by different names such as Aquaporin-5 and sometimes referenced in studies as Aquazol 5.

Aquaporins form homotetrameric transmembrane channels, with each monomer independently mediating water transport across the plasma membrane along its osmotic gradient (PubMed : 18768791, PubMed : 8621489). Plays an important role in fluid secretion in salivary glands (By similarity). Required for TRPV4 activation by hypotonicity. Together with TRPV4, controls regulatory volume decrease in salivary epithelial cells (PubMed : 16571723).
