The ARAP1 protein also known as Centaurin delta-1 plays a role in mechanics acting as an ArfGAP with RhoGAP domain and Ankyrin repeat protein. ARAP1 weighs approximately 150 kDa and mainly expresses in tissues including the heart liver and skeletal muscle. This protein localizes primarily in the cytoplasm and is involved in cellular processes by regulating phosphoinositide 3-kinase (PI3K) signaling.
ARAP1 coordinates the remodeling of the actin cytoskeleton. It assists in cell adhesion migration and endocytosis. Through its GAP domains ARAP1 regulates small GTPases such as Arf and Rho proteins which participate in vesicular trafficking and cytoskeletal dynamics. While not a member of a specific complex it interacts with signaling molecules to influence cellular shape and movement.
ARAP1 acts in the PI3K/AKT and Rho signaling pathways shaping cellular responses to external stimuli. It regulates the localization and activity of phosphoinositide phosphates important for the PI3K/AKT pathway. ARAP1's interaction with proteins like Akt and RhoA affects their signaling to produce changes in cell motility and growth integrating signals from various upstream activators.
ARAP1 has links to cancer and type 2 diabetes. In cancer ARAP1 may contribute to tumor cell invasiveness relating to its function in actin cytoskeleton regulation and its interaction with RhoA. Additionally its association with PI3K/AKT pathway dysregulation connects ARAP1 to insulin signaling disruption relevant in type 2 diabetes. Understanding these interactions may provide insights into therapeutic strategies.
