ARF1 and ARF3 also known as ADP-ribosylation factors 1 and 3 are small GTP-binding proteins belonging to the ARF family. ARF1 has a molecular mass of approximately 21 kDa. These proteins are expressed in most eukaryotic cells where they play roles in vesicular trafficking processes. ARF1 is mainly located in the Golgi apparatus while ARF3 appears in the plasma membrane and cytoplasm. They function as molecular switches by cycling between active GTP-bound and inactive GDP-bound states which regulates the formation and budding of vesicles.
Biological function summary

ARF1 and ARF3 are significant in the intracellular trafficking of proteins and lipids. The proteins are part of the coatomer protein complex (COPI) important for retrograde transport from the Golgi to the endoplasmic reticulum. By recruiting coat proteins to the Golgi membranes ARF1 and ARF3 facilitate the sorting and packaging of cargo molecules into transport vesicles. This process sustains the maintenance of organelle identity and the dynamics of membrane remodeling.

Pathways

ARF1 and ARF3 are involved in key cellular pathways particularly those related to vesicular trafficking and membrane dynamics. They participate in the COPI and clathrin-mediated pathways. Through these pathways ARF1 interacts with proteins like PDZD8 while ARF3 works with proteins involved in primary cilia formation. These interactions underline their integrative role in maintaining proper cellular organization and communication.

ARF1 and ARF3 have links to cancer and neurodegenerative diseases. Dysregulation of ARF1 is associated with various cancer types due to its role in cell proliferation and migration. ARF3 has connections to neurodegenerative disorders where altered vesicular trafficking can lead to pathological conditions. ARF1's interaction with the protein NFDM-442 highlights its importance in disease contexts whereby changes in these interactions might contribute to disease progression and severity.

Small GTPase involved in protein trafficking between different compartments (PubMed : 8253837). Modulates vesicle budding and uncoating within the Golgi complex (PubMed : 8253837). In its GTP-bound form, triggers the recruitment of coatomer proteins to the Golgi membrane (PubMed : 8253837). The hydrolysis of ARF1-bound GTP, which is mediated by ARFGAPs proteins, is required for dissociation of coat proteins from Golgi membranes and vesicles (PubMed : 8253837). The GTP-bound form interacts with PICK1 to limit PICK1-mediated inhibition of Arp2/3 complex activity; the function is linked to AMPA receptor (AMPAR) trafficking, regulation of synaptic plasticity of excitatory synapses and spine shrinkage during long-term depression (LTD) (By similarity). Plays a key role in the regulation of intestinal stem cells and gut microbiota, and is essential for maintaining intestinal homeostasis (By similarity).
