Supplementary information

ARHGAP1 also known as Rho GTPase-activating protein 1 plays an important role in the regulation of the Rho family of GTPases. These proteins control the actin cytoskeleton affecting cell shape and migration. ARHGAP1 acts as a GTPase-activating protein stimulating the GTPase activity of Rho-type GTPases and thereby controls their signaling activity. The protein weighs about 48 kDa and shows expression in various tissues including lung heart and liver.
Biological function summary

ARHGAP1 contributes to cell movement processes such as migration and invasion. It is part of a regulatory complex that modulates the signaling pathways involving actin cytoskeletal dynamics. ARHGAP1 interacts with key molecules in pathways that manage cell adhesion and motility indicating its role in cellular dynamics and structural integrity.

Pathways

ARHGAP1 actively participates in the Rho GTPase cycle interacting with proteins like Cdc42 and Rac which are involved in controlling actin polymerization dynamics. The protein plays a role in pathways like the VEGF signaling pathway influencing how cells respond to growth signals. It provides an intersection between small GTPase signaling and cytoskeletal rearrangement essential for cell movement and stability.

ARHGAP1 shows associations with cancer progression and metastasis. Changes in ARHGAP1 expression can influence the behavior of cancer cells increasing their migratory and invasive capabilities. In addition ARHGAP1 connects with the protein EGFR where aberrant pathways involving these proteins can lead to non-small cell lung cancer. In this context it is important to further investigate ARHGAP1's roles to understand its potential use in therapeutic developments.

Target data

GTPase activator for the Rho, Rac and Cdc42 proteins, converting them to the putatively inactive GDP-bound state. Cdc42 seems to be the preferred substrate.
