ARHGEF3 also known as Rho guanine nucleotide exchange factor 3 functions in the regulation of Rho GTPases by accelerating the GDP-GTP exchange. ARHGEF3 weighs approximately 105 kDa and plays a significant role in cellular processes that require cytoskeletal reorganization. It is broadly expressed in various tissues particularly within hematopoietic cells and the bone marrow.
The regulatory role of ARHGEF3 impacts the actin cytoskeleton which is fundamental for cell shape and movement. ARHGEF3 activates RhoA and other Rho GTPases which are important for the formation of stress fibers and focal adhesions. As part of a signaling complex ARHGEF3 interacts with several adaptor proteins to transmit extracellular signals to intracellular responses.
ARHGEF3 integrates into the Rho signaling pathway and the Wnt signaling pathway. It collaborates with proteins like ROCK1 and CDC42 that mediate various downstream effects resulting in cytoskeletal arrangement and cellular motility. These interactions highlight its role in facilitating dynamic cellular changes essential for various physiological responses.
ARHGEF3 has relevance to osteoporosis and anemia. Variations in ARHGEF3 expression influence bone density and are investigated for potential links to osteoporosis. Additionally its expression levels affect erythrocyte maturation connecting ARHGEF3 to anemia. In these contexts ARHGEF3 interacts indirectly with proteins such as RANKL in bone metabolism and EPO in erythropoiesis.
