Supplementary information

The ARID3A protein also known as AT-rich interaction domain 3A functions mechanically as a DNA binding protein involved in transcriptional regulation. It contains an AT-rich interaction domain and acts as a transcription factor. The mass of ARID3A is approximately 63 kDa. This protein is expressed in various tissues with notable expression in B lymphocytes. It plays a role in modulating gene expression by binding to specific DNA sequences.
Biological function summary

ARID3A regulates distinct processes in cell differentiation and development. This protein is essential for normal B cell development and function facilitating the transition of B cells through various stages of maturation. ARID3A does not operate in isolation; it forms part of larger protein complexes that interact with other proteins to regulate transcription. By doing so it ensures specific gene expression patterns necessary for correct cell lineage determination.

Pathways

Multiple cellular systems necessitate ARID3A's involvement. It acts within the B cell receptor (BCR) signaling pathway where it collaborates with other proteins such as BLNK to promote B cell responses. ARID3A also participates in the cell cycle regulation pathway influencing cell cycle progression by directly affecting the transcription of genes that regulate cell division.

ARID3A is implicated in the pathogenesis of certain autoimmune conditions like systemic lupus erythematosus (SLE). Researchers observed differences in expression levels of ARID3A in patients with SLE linking its dysregulation to disease activity. Additionally ARID3A has connections to hematological malignancies with leukemia and lymphoma cells often showing altered ARID3A expression. Proteins such as RAG1 which are connected to ARID3A through these diseases also exhibit dysregulation further affecting B cell function.
