Function

Transcription coactivator that binds to the 5'-AATA[CT]-3' core sequence and plays a key role in adipogenesis and liver development. Acts by forming a complex with phosphorylated PHF2, which mediates demethylation at Lys-336, leading to target the PHF2-ARID5B complex to target promoters, where PHF2 mediates demethylation of dimethylated 'Lys-9' of histone H3 (H3K9me2), followed by transcription activation of target genes. The PHF2-ARID5B complex acts as a coactivator of HNF4A in liver. Required for adipogenesis: regulates triglyceride metabolism in adipocytes by regulating expression of adipogenic genes. Overexpression leads to induction of smooth muscle marker genes, suggesting that it may also act as a regulator of smooth muscle cell differentiation and proliferation. Represses the cytomegalovirus enhancer.

Activity summary

ARID5B also known as AT-rich interactive domain-containing protein 5B functions as a transcription factor. This protein with a mass of approximately 132 kilodaltons interacts with DNA to regulate gene expression by binding to specific AT-rich sequences. ARID5B localizes in the nucleus and exhibits expression in various tissues including the lungs spleen and liver highlighting its involvement in multiple biological processes.

Biological function summary

The activities of AT-rich sequences within the gene regulatory network depend significantly on transcription factors. ARID5B plays an important role in modulating the expression of genes involved in cell differentiation and development. It functions within a complex of proteins that participate in chromatin remodeling which alters the accessibility of chromatin to the transcriptional machinery. Through these interactions ARID5B ensures precise control over gene expression necessary for proper cellular function and organism development.

Pathways

Transcription factors like ARID5B integrate signals from various regulatory networks. ARID5B engages in the Wnt signaling and Notch pathways influencing stem cell differentiation and maintaining tissue homeostasis. In these pathways ARID5B interacts with other proteins like TCF/LEF transcription factors in the Wnt pathway and NICD (Notch intracellular domain) in the Notch pathway coordinating the transcriptional response required for cell fate determination.

Associated diseases and disorders

ARID5B mutations or dysregulation have associations with certain types of cancer and autoimmune diseases. Acute lymphoblastic leukemia (ALL) shows links to altered ARID5B function contributing to abnormal cell proliferation. Additionally variations in ARID5B influence the risk of developing type 1 diabetes where it interacts with other immune-related proteins such as HLA-DRB1. Researchers study these interactions to understand disease mechanisms and explore potential therapeutic strategies.
