ARIH1 also known as Human Ubiquitin-Protein Ligase ARIH1 is a 55-kDa protein involved in the ubiquitination process. Ubiquitination is a process that tags proteins for degradation. ARIH1 functions as an E3 ubiquitin-protein ligase facilitating the transfer of ubiquitin to specific substrate proteins. It acts with ubiquitin-conjugating enzymes mediating the attachment of ubiquitin molecules to protein targets. ARIH1 is mainly expressed in the cytoplasm and nucleus of cells with higher expression detected in the brain and testes.
Biological function summary

ARIH1 plays a role in the regulation of several cellular processes such as protein degradation signal transduction and DNA repair. It participates in the formation of the ARIH1-E2 ubiquitin-protein ligase complex which modulates protein stability by marking them for proteasomal degradation. This complex ensures the controlled turnover of key regulatory proteins to maintain cellular homeostasis. ARIH1 also influences the function of other important molecules through its ubiquitination activity impacting cellular responses to stress.

Pathways

ARIH1 gets involved in the ubiquitin-proteasome system which orchestrates the degradation of unneeded or damaged proteins. ARIH1 operates within this pathway interacting with proteins like the E2 conjugating enzyme UBE2L3. Moreover it participates in the NEDDylation pathway which adds another layer of regulation to protein stability and function. Within these pathways ARIH1 ensures effective control over multiple cellular mechanisms thereby supporting cellular function.

ARIH1 associates with conditions like cancer and neurodegenerative diseases. In cancer altered ubiquitination processes involving ARIH1 may disrupt normal cellular regulation contributing to tumorigenesis. Additionally ARIH1's dysregulation impacts neurodegenerative diseases potentially linked to improper protein degradation in neurons. ARIH1's interactions with proteins such as p53 and PARK2 further connect it to these disorders highlighting its role in the pathological processes.

E3 ubiquitin-protein ligase, which catalyzes ubiquitination of target proteins together with ubiquitin-conjugating enzyme E2 UBE2L3 (PubMed : 15236971, PubMed : 21532592, PubMed : 23707686, PubMed : 24076655, PubMed : 27565346). Acts as an atypical E3 ubiquitin-protein ligase by working together with cullin-RING ubiquitin ligase (CRL) complexes and initiating ubiquitination of CRL substrates : associates with CRL complexes and specifically mediates addition of the first ubiquitin on CRLs targets (PubMed : 27565346). The initial ubiquitin is then elongated by CDC34/UBE2R1 and UBE2R2 (PubMed : 27565346). According to a report, EIF4E2 ubiquitination leads to promote EIF4E2 cap-binding and protein translation arrest (PubMed : 25624349). According to another report EIF4E2 ubiquitination leads to its subsequent degradation (PubMed : 14623119). Acts as the ligase involved in ISGylation of EIF4E2 (PubMed : 17289916). In vitro, controls the degradation of the LINC (LInker of Nucleoskeleton and Cytoskeleton) complex member SUN2 and may therefore have a role in the formation and localization of the LINC complex, and as a consequence, nuclear subcellular localization and nuclear morphology (PubMed : 29689197).
