Activity summary

ARL8A and ARL8B also known as ADP-ribosylation factor-like protein 8A and 8B are small GTPases with masses of approximately 21 kDa. They belong to the ARF-like GTPase family and they localize mainly on lysosomes and late endosomes. Both ARL8A and ARL8B widely express in tissues but show higher levels in certain types like immune cells. Their structural and functional similarities allow them to share roles in cellular functions related to lysosomal positioning and movement.

Biological function summary

ARL8A and ARL8B regulate the anterograde transport of lysosomes and are part of the lysosomal trafficking complex. They help in organizing lysosomal positioning a role important for normal lysosome function. This function includes interaction and formation of a complex with other proteins such as PLEKHM2 and SKIP which facilitate the attachment of lysosomes to microtubule tracks via kinesin motor proteins. This movement allows lysosomes to fulfill their roles in cellular distribution and degradation processes.

Pathways

ARL8A and ARL8B play roles in the lysosomal protein transport pathway and autophagy. They engage in lysosomal motility impacting autophagic flux a process essential for cellular homeostasis and stress response. ARL8B in particular interacts with the HOPS complex promoting fusion between autophagosomes and lysosomes therefore influencing autophagy. Among related proteins Rab7A is noteworthy as a regulator of late endosomal trafficking pathways that interface with processes involving ARL8 proteins.

Associated diseases and disorders

ARL8A and ARL8B are associated with neurodegenerative diseases and cancer. Dysregulation in lysosomal positioning can impair autophagy linking these proteins to neurodegenerative conditions like Parkinson's disease. In cancer altered lysosomal trafficking influences tumor cell invasion and metastasis. Both proteins interact with LAMP proteins which contribute to the lysosomal membrane integrity central to diseases involving lysosomal dysfunction.
