Actin-binding component of the Arp2/3 complex, a multiprotein complex that mediates actin polymerization upon stimulation by nucleation-promoting factor (NPF) (PubMed:9230079). The Arp2/3 complex mediates the formation of branched actin networks in the cytoplasm, providing the force for cell motility (PubMed:9230079). In addition to its role in the cytoplasmic cytoskeleton, the Arp2/3 complex also promotes actin polymerization in the nucleus, thereby regulating gene transcription and repair of damaged DNA (PubMed:29925947). The Arp2/3 complex promotes homologous recombination (HR) repair in response to DNA damage by promoting nuclear actin polymerization, leading to drive motility of double-strand breaks (DSBs) (PubMed:29925947).

The ARPC4 protein also known as Actin-Related Protein 2/3 Complex Subunit 4 plays a significant role in the actin nucleation process. It is a component of the ARP2/3 complex which is essential for the formation of branched actin filament networks. ARPC4 has a molecular weight of approximately 20 kDa. It is ubiquitously expressed across various tissues indicating its fundamental role in cellular mechanisms.
ARPC4 contributes to the dynamic remodeling of the actin cytoskeleton. It is an integral part of the ARP2/3 complex a multi-protein structure that facilitates actin filament branching promoting cell movement and shape changes. This complex is critical for processes like cell migration phagocytosis and intracellular trafficking. Lane 1: HeLa whole cell lysate at 15 µg
Lane 2: 293T whole cell lysate at 15 µg
