ARS2 also known as SRRT (serrate RNA effector molecule homolog) is a human protein encoded by the SRRT gene. ARS2 is a nuclear protein with an approximate molecular weight of 96 kDa. It is highly expressed in tissues such as the brain lung and liver. Inside the nucleus ARS2 interacts with various RNA-processing complexes binding to nascent RNA transcripts. The primary role of ARS2 centers around RNA metabolism where it manages and organizes the maturation of small RNAs.
Biological function summary

This protein assists in the capping processing and export of several RNA types including pre-miRNAs and other short-length RNAs. ARS2 forms part of a larger protein complex interacting with the Cap Binding Complex (CBC) and Integrator complex which helps regulate RNA stability and processing. Through these interactions ARS2 facilitates the proper maturation and quality control of RNA which is essential for maintaining cellular function and gene expression regulation.

Pathways

The activity of this protein notably aligns with the mRNA processing pathway and RNA surveillance pathway. Within these pathways ARS2 plays a role by participating in the post-transcriptional regulation that is interlinked with proteins like CBC and the nuclear RNA exosome complex. These collaborations enable tight control over RNA homeostasis ensuring that cells can adapt and respond to various cellular signals and stressors by modifying gene expression profiles.

Abnormalities in ARS2 function associate with neural development issues and cancer. Acts as a mediator between the cap-binding complex (CBC) and the primary microRNAs (miRNAs) processing machinery during cell proliferation. Contributes to the stability and delivery of capped primary miRNA transcripts to the primary miRNA processing complex containing DGCR8 and DROSHA, thereby playing a role in RNA-mediated gene silencing (RNAi) by miRNAs. Binds capped RNAs (m7GpppG-capped RNA); however interaction is probably mediated via its interaction with NCBP1/CBP80 component of the CBC complex. Involved in cell cycle progression at S phase. Does not directly confer arsenite resistance but rather modulates arsenic sensitivity. Independently of its activity on miRNAs, necessary and sufficient to promote neural stem cell self-renewal. Does so by directly binding SOX2 promoter and positively regulating its transcription (By similarity).
