Artemin also known as ART or ARTN is a neurotrophic factor and part of the glial cell line-derived neurotrophic factor (GDNF) family. It comprises a mature peptide with a molecular mass of approximately 24 kDa. This protein expresses highly in the peripheral and central nervous system particularly in developing and injured tissues. Artemin supports the survival and differentiation of specific neurons contributing to neurite outgrowth and neuronal regeneration.
Biological function summary

Artemin is a significant modulator of neuronal survival and neurite extension. It forms a signaling complex with its receptor components the GDNF family receptor alpha 3 (GFRα3) and the receptor tyrosine kinase (RET). This complex initiates signaling cascades essential for embryonic neuronal development and adult neuroprotection. Artemin selectively activates certain pathways that promote neuronal survival and regenerative responses.

Pathways

Artemin engages with the phosphatidylinositol 3-kinase/Protein Kinase B (PI3K/AKT) and the mitogen-activated protein kinase (MAPK/ERK) pathways. These pathways are critical for cell survival proliferation and differentiation. Artemin closely associates with other neurotrophic factors such as GDNF within these pathways suggesting a shared or complementary role in neuroprotection.

Artemin shows potential involvement in neurodegenerative diseases and peripheral neuropathies. It may have therapeutic implications in conditions such as Parkinson's disease where it enhances neuronal survival. Similarly its role in peripheral neuropathy suggests that it modulates pain through interactions with receptors like GFRα3 possibly impacting RET signaling pathways.

Growth factor that supports the survival of sensory and sympathetic peripheral neurons in culture and also supports the survival of dopaminergic neurons of the ventral mid-brain (PubMed : 10583383, PubMed : 9883723). Acts by binding to its coreceptor, GFRA3, leading to autophosphorylation and activation of the RET receptor (PubMed : 31535977). Strong attractant of gut hematopoietic cells thus promoting the formation Peyer's patch-like structures, a major component of the gut-associated lymphoid tissue (By similarity).
