Immunofluorescence analysis of GFRA3-Ret51 receptor complex bound Artemin. Human U2OS cells were transfected with GFRa3-Ret51 expression vector followed by treatment with 100 nM Artemin. Artemin bound to the GFRa3-Ret51 receptor complex was detected with ab205564 at 20 μg/mL. Goat anti-mouse Dylight 488 conjugated antibody (1 μg/mL) was used as secondary antibody. Biological function summary
Artemin is a significant modulator of neuronal survival and neurite extension. It forms a signaling complex with its receptor components the GDNF family receptor alpha 3 (GFRα3) and the receptor tyrosine kinase (RET). This complex initiates signaling cascades essential for embryonic neuronal development and adult neuroprotection. Artemin selectively activates certain pathways that promote neuronal survival and regenerative responses.
Pathways
Artemin engages with the phosphatidylinositol 3-kinase/Protein Kinase B (PI3K/AKT) and the mitogen-activated protein kinase (MAPK/ERK) pathways. These pathways are critical for cell survival proliferation and differentiation. Artemin closely associates with other neurotrophic factors such as GDNF within these pathways suggesting a shared or complementary role in neuroprotection.
