Immunohistochemistry (Formalin/PFA-fixed paraffin-embedded sections) analysis of human bladder carcinoma tissue sections labeling ARTS1/ERAP1 with Purified ab124669 at 1 : 1000 dilution. Heat mediated antigen retrieval was performed using ab93684 (Tris/EDTA buffer, pH 9.0). Tissue was counterstained with Hematoxylin. ImmunoHistoProbe one step HRP Polymer (ready to use) secondary antibody was used at 1 : 0 dilution. PBS instead of the primary antibody was used as the negative control.

ARTS1 also known as ERAP1 (Endoplasmic Reticulum Aminopeptidase 1) is an aminopeptidase enzyme with a molecular mass of about 110 kDa. This protein is located in the endoplasmic reticulum and plays a significant mechanical role in trimming peptides for presentation by MHC class I molecules. Its expression is highly detectable in cells with robust antigen-presenting capabilities such as dendritic cells and macrophages. Additionally ARTS1/ERAP1 exists in other tissues where immune surveillance is essential.
Biological function summary

The function of ARTS1/ERAP1 involves the processing of proteolytic fragments preparing them for optimal binding to MHC class I molecules. This enzyme participates in a complex alongside ERAP2 another aminopeptidase working cooperatively to adjust peptide lengths to the correct size necessary for MHC class I presentation. Through this role ARTS1/ERAP1 influences the immune system's ability to detect and respond to infected or neoplastic cells by ensuring that antigenic peptides are properly presented.

Pathways

ARTS1/ERAP1 is a critical component of the antigen processing and presentation pathway. This pathway is essential for the presentation of endogenous antigens to cytotoxic T lymphocytes therefore playing a role in adaptive immunity. Related proteins include MHC class I molecules which present the processed peptides and TAP (Transporter associated with Antigen Processing) which transports peptides into the endoplasmic reticulum for further trimming by ARTS1/ERAP1.

Research links ARTS1/ERAP1 to ankylosing spondylitis and other autoimmune diseases. Variations in the ERAP1 gene may affect peptide trimming efficiency altering the peptide repertoire presented by MHC class I molecules and influencing autoimmune activation. The link to another protein HLA-B27 is notable in ankylosing spondylitis as this MHC class I allele frequently associates with the disease highlighting a complex interaction between ARTS1/ERAP1 enzymatic function and autoimmune susceptibility.

Aminopeptidase that plays a central role in peptide trimming, a step required for the generation of most HLA class I-binding peptides. Peptide trimming is essential to customize longer precursor peptides to fit them to the correct length required for presentation on MHC class I molecules. Strongly prefers substrates 9-16 residues long. Rapidly degrades 13-mer to a 9-mer and then stops. Preferentially hydrolyzes the residue Leu and peptides with a hydrophobic C-terminus, while it has weak activity toward peptides with charged C-terminus. May play a role in the inactivation of peptide hormones. May be involved in the regulation of blood pressure through the inactivation of angiotensin II and/or the generation of bradykinin in the kidney.
