Supplementary information

ARVCF also known as Armadillo Repeat gene deleted in Velo-Cardio-Facial Syndrome encodes a protein involved in cell adhesion and signaling. This protein weighs around 86 kDa and contains armadillo repeat domains facilitating interactions with other membrane-associated proteins. ARVCF is expressed in various tissues including the heart and brain suggesting its versatile role in cellular functioning. Localization in adherens junctions indicates its importance in maintaining structural integrity.
Biological function summary

The function of ARVCF supports cell adhesion and communication. It participates in complex formation with catenins and cadherins which are critical for adherens junctions. These complexes enable the anchoring of cytoskeletal elements to the cell membrane aiding in stable cell-cell contact. ARVCF modulates the transduction of signals that regulate cellular behaviors such as migration and proliferation through these interactions.

Pathways

ARVCF plays a role in Wnt signaling and adherens junction pathways. The protein interacts with β-catenin a significant player in Wnt pathway influencing gene expression related to cell growth and differentiation. ARVCF also associates with proteins involved in the assembly and maintenance of adherens junctions further emphasizing its connection to cellular architecture and signaling.

ARVCF has been linked to velo-cardio-facial syndrome and some forms of cancer. These associations suggest a connection between ARVCF dysfunction and disruptions in normal cell signaling and adhesion processes. In cancer altered ARVCF expression can impact the interaction with β-catenin and other junction-related proteins potentially influencing tumor progression and metastasis.
